Effect of age and season on some semen parameters of Nili-Ravi buffalo (Bubalus bubalis) bulls
نویسندگان
چکیده
This study was carried out on 16 buffalo bulls (Bubalus bubalis) divided into four age groups, including three healthy (less than 5, 6-10, and more than 11 year old animals), and one abnormal group (6-10 year old bulls having poor semen quality). The study was undertaken for a one year period divided into five seasons (dry summer, humid summer, autumn, winter and spring). Overall semen volume was 4.67±1.62 ml and showed a nonsignificant difference between age groups, although it was higher (P<0.05) in autumn. Overall semen pH was 6.55±0.50 and was lower (P<0.05) in the 6-10 than in the 11 year old bulls and the abnormal group. It was lower (P<0.05) in autumn while higher (P<0.05) in winter. Colour score was higher (P<0.05) in less than 5 year old bulls. However, it was lower (P<0.05) in the abnormal group than in healthy groups but was lower (P<0.05) in dry summer. Overall mass activity score was 2.65±1.03 and was lower (P<0.05) in more than 11 year old bulls. However, it was lower (P<0.05) in the abnormal than in healthy groups. Mass activity score was higher (P<0.05) in dry summer and spring. Overall sperm motility was 56.89±0.65% and showed no difference in healthy groups, but was lower (P<0.05) in the abnormal group. It was lower (P<0.05) in winter than humid summer and autumn. Overall sperm concentration was 1.00±0.50 millions/ml and was higher (P<0.05) in less than 5 year old bulls. However, it was lower (P<0.05) in the abnormal group. Sperm concentration was lower (P<0.05) in winter. The pH of semen showed negative (P<0.001) correlation with colour (r=-0.38), sperm concentration (r=-0.37), mass activity (r=-0.37) and motility (r=0.17). Sperm concentration correlated positively (P<0.001) with mass activity (r=0.86) and progressive motility (r=0.16).

Assessment of Nili-Ravi buffalo (Bubalus bubalis) semen by MTT reduction assay
MTT (3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay is commonly used to validate the viability of metabolically active cells. The study was conducted to examine and validate the MTT test to assess the sperm viability of Nili-Ravi buffalo bulls and compare the efficiency of the test with the supra-vital staining technique (eosin-nigrosine) and hypo-osmotic swelling test. متن کاملReproductive characteristics and thyroidal function in relation with season in Khuzestan buffalo (Bubalus bubalis) bulls
High ambient temperature is the major constraint on Buffalo productivity. The aim of this study was to evaluate the reproductive performance and thyroid gland function in winter and summer seasons in Khuzestan buffalo bulls. Six male indigenous buffaloes of Khuzestan with nearly the same age (2-3 years old) and weight were used. Semen and blood samples through jugular vein were collected, every...
متن کاملEffect of L-cysteine on Post Thaw Quality of Nili Ravi Buffalo (bubalus Bubalis) Bull Spermatozoa Cryopreserved in Tris Citric Acid Extender
This study was designed to determine effect of various concentrations of Lcysteine on frozen-thaw semen quality of Nili Ravi buffalo. Single ejaculate of four buffalo bulls (Nili-Ravi) was collected at weekly intervals for 5 weeks. Ejaculates of acceptable motility (≥70%) were pooled and semen was diluted by tris citric acid extender (TCAE; 300 mosm/kg osmotic pressure and ~7.0 pH) having 4 lev...
متن کاملThe in vitro effect of leptin on semen quality of water buffalo (Bubalus bubalis) bulls
The purpose of this study was to evaluate the probable effects of leptin addition in different levels to the semen extender on sperm quality (motility and motility parameters, viability, sperm membrane integrity, and DNA damage). Semen specimens were evaluated immediately after leptin addition, equilibration time and after thawing the frozen semen. Five healthy buffalo bulls (5 ejaculates from ...
متن کاملP-103: Effects of In Vitro Selenium Supplementation on Buffalo (Bubalus bubalis) Sperm Quality following Cryopreservation
Background: Artificial insemination is a valuable tool in genetic improvement programs and widely used breeding technique in farm animals. Semen processing and cryopreservation causes considerable damage to sperm DNA, motility apparatus, plasma membrane, acrosomal cap and thus, reduced fertility. Selenium is an integral part of glutathione peroxidase (GSH-Px), an enzyme which protects cell inte...
